Kỹ thuật di truyền hay kỹ thuật gen là thao tác thay đổi gen bằng công nghệ sinh học. Nó bao gồm các phương pháp kỹ thuật dùng để thay đổi thành phần di truyền của các tế bào, bao gồm sự dịch chuyển gen cùng loài và khác loài để tạo ra những sinh vật mới hoặc hoàn hảo hơn. Kỹ thuật di truyền là một quá trình làm thay đổi cấu trúc di truyền của một sinh vật bằng cách loại bỏ hoặc đưa vào DNA. Không giống như nhân giống động vậtthực vật truyền thống, bao gồm thực hiện nhiều phép lai và sau đó chọn ra sinh vật có kiểu hình mong muốn, kỹ thuật di truyền lấy gen trực tiếp từ sinh vật này và chuyển nó sang sinh vật khác. Điều này nhanh hơn nhiều, có thể được sử dụng để chèn bất kỳ gen nào từ bất kỳ sinh vật nào (ngay cả những gen từ các miền khác nhau) và ngăn chặn các gen không mong muốn khác cũng được thêm vào.[3]

Thực vật, động vật hoặc vi sinh vật đã bị thay đổi thông qua kỹ thuật di truyền được gọi là sinh vật biến đổi gen hoặc GMO.[14] Nếu vật liệu di truyền từ loài khác được thêm vào vật chủ, sinh vật tạo thành được gọi là chuyển gen. Lịch sử

Con người đã thay đổi bộ gen của các loài trong hàng nghìn năm thông qua chọn lọc giống, hoặc chọn lọc nhân tạo [20] :1 [21] :1 trái ngược với chọn lọc tự nhiên. Gần đây hơn, nhân giống đột biến đã sử dụng tiếp xúc với hóa chất hoặc bức xạ để tạo ra một tần số cao các đột biến ngẫu nhiên, cho các mục đích chọn lọc. Năm 1972, Paul Berg đã tạo ra các phân tử DNA tái tổ hợp đầu tiên bằng cách kết hợp DNA của virus khỉ SV40 với DNA của virus lambda.[26] Năm 1973, Herbert BoyerStanley Cohen đã tạo ra sinh vật chuyển gen đầu tiên bằng cách đưa gen kháng thuốc kháng sinh vào plasmid của vi khuẩn Escherichia coli.[27][28] Một năm sau, Rudolf Jaenisch đã tạo ra một con chuột chuyển gen bằng cách đưa DNA ngoại lai vào phôi thai của nó, biến nó trở thành động vật chuyển gen đầu tiên trên thế giới [29] Những thành tựu này đã dẫn đến mối quan tâm trong cộng đồng khoa học về những rủi ro tiềm ẩn từ kỹ thuật di truyền, lần đầu tiên được thảo luận sâu tại Hội nghị Asilomar năm 1975. Genentech công bố sản xuất insulin người được biến đổi gen vào năm 1978.[32] Năm 1980, Tòa án Tối cao Hoa Kỳ trong vụ án Diamond v. Chakrabaty phán quyết rằng sinh vật bị biến đổi gen có thể được cấp bằng sáng chế.[33] Insulin do vi khuẩn sản xuất đã được Cục Quản lý Thực phẩm và Dược phẩm (FDA) chấp thuận cho phát hành vào năm 1982.[34]

Năm 1983, một công ty công nghệ sinh học, Advanced Genetic Sciences (AGS) đã xin phép chính phủ Hoa Kỳ thực hiện các thử nghiệm thực địa với chủng Pseudomonas syringae trừ băng để bảo vệ cây trồng khỏi sương giá, nhưng các nhóm môi trường và những người phản đối đã trì hoãn các thử nghiệm trên thực địa trong bốn năm với thách thức pháp lý.[35] Năm 1987, chủng vi khuẩn P. syringae trừ băng trở thành sinh vật biến đổi gen (GMO) đầu tiên được phát tán vào môi trường [36] khi ruộng dâu tây và ruộng khoai tây ở California bị phun thuốc.[37] Cả hai cánh đồng thử nghiệm đã bị tấn công bởi các nhóm hoạt động vào đêm trước khi các cuộc thử nghiệm xảy ra: "Khu thử nghiệm đầu tiên trên thế giới đã thu hút kẻ phá ruộng đầu tiên trên thế giới".[36]

Các thử nghiệm đầu tiên trên thực địa đối với cây biến đổi gen diễn ra ở Pháp và Mỹ vào năm 1986, cây thuốc lá đã được biến đổi gen để kháng thuốc diệt cỏ.[38] Cộng hòa Nhân dân Trung Hoa là nước đầu tiên thương mại hóa cây chuyển gen, giới thiệu một loại thuốc lá kháng vi rút vào năm 1992.[39] Năm 1994, Calgene được chấp thuận phát hành thương mại loại thực phẩm biến đổi gen đầu tiên, Flavr Savr, một loại cà chua được thiết kế để có thời hạn sử dụng lâu hơn.[40] Năm 1994, Liên minh Châu Âu đã phê duyệt thuốc lá được biến đổi gen để kháng thuốc diệt cỏ bromoxynil, biến nó trở thành cây trồng biến đổi gen đầu tiên được thương mại hóa ở Châu Âu.[41] Năm 1995, Khoai tây Bt đã được Cơ quan Bảo vệ Môi trường phê duyệt an toàn, sau khi được FDA phê duyệt, trở thành cây trồng sản xuất thuốc trừ sâu đầu tiên được chấp thuận ở Mỹ.[42] Năm 2009, 11 cây chuyển gen đã được trồng đại trà ở 25 quốc gia, trong đó lớn nhất theo diện tích được trồng là Mỹ, Brazil, Argentina, Ấn Độ, Canada, Trung Quốc, Paraguay và Nam Phi.[43]

Năm 2010, các nhà khoa học tại Viện J. Craig Venter đã tạo ra bộ gen tổng hợp đầu tiên và đưa nó vào một tế bào vi khuẩn trống. Tạo ra một GMO là một quá trình gồm nhiều bước. Các kỹ sư di truyền trước tiên phải chọn loại gen mà họ muốn đưa vào sinh vật. Tế bào chứa gen được mở ra và DNA được tinh sạch.[53] Gen được tách ra bằng cách sử dụng các enzym giới hạn để cắt ADN thành các đoạn [54] hoặc phản ứng chuỗi polymerase (PCR) để khuếch đại đoạn gen.[55] Các phân đoạn này sau đó có thể được chiết xuất thông qua điện di trên gel. Nếu gen được chọn hoặc bộ gen của sinh vật hiến tặng đã được nghiên cứu kỹ lưỡng thì có thể đã có thể truy cập được từ thư viện di truyền. Nếu trình tự DNA được biết, nhưng không có bản sao của gen, nó cũng có thể được tổng hợp nhân tạo.[56] Một khi đã phân lập được gen được ligated vào một plasmid mà sau đó được đưa vào một loại vi khuẩn. Plasmid được sao chép khi vi khuẩn phân chia, đảm bảo có sẵn các bản sao không giới hạn của gen.[57]

Trước khi gen được đưa vào sinh vật mục tiêu, nó phải được kết hợp với các yếu tố di truyền khác. Chúng bao gồm vùng khởi động và vùng kết thúc, khởi đầu và kết thúc phiên mã. Một gen đánh dấu có thể lựa chọn được thêm vào, trong hầu hết các trường hợp, gen này tạo ra khả năng kháng thuốc kháng sinh, vì vậy các nhà nghiên cứu có thể dễ dàng xác định tế bào nào đã được biến nạp thành công. Gen cũng có thể được sửa đổi ở giai đoạn này để biểu hiện hoặc hiệu quả tốt hơn. Những thao tác này được thực hiện bằng kỹ thuật DNA tái tổ hợp, chẳng hạn như tiêu hóa giới hạn, nối và nhân bản phân tử.[58]

Có một số kỹ thuật được sử dụng để chèn vật liệu di truyền vào bộ gen của vật chủ. Một số vi khuẩn có thể tiếp nhận DNA ngoại lai một cách tự nhiên. Khả năng này có thể được tạo ra ở các vi khuẩn khác thông qua căng thẳng (ví dụ như sốc nhiệt hoặc điện), làm tăng tính thấm của màng tế bào đối với DNA; DNA bổ sung có thể tích hợp với bộ gen hoặc tồn tại dưới dạng DNA ngoài nhiễm sắc thể. DNA thường được đưa vào tế bào động vật bằng cách sử dụng vi tiêm, nơi nó có thể được tiêm trực tiếp qua vỏ nhân của tế bào vào nhân hoặc thông qua việc sử dụng các vectơ virut.[59]

Bộ gen thực vật có thể được thiết kế bằng các phương pháp vật lý hoặc bằng cách sử dụng vi khuẩn Agrobacterium để phân phối các trình tự được lưu trữ trong vectơ nhị phân T-DNA. Ở thực vật, DNA thường được chèn bằng cách sử dụng phương pháp biến nạp qua Agrobacterium,[60] tận dụng trình tự T-DNA của vi khuẩn Agrobacterium cho phép đưa vật liệu di truyền vào tế bào thực vật một cách tự nhiên.[61] Các phương pháp khác bao gồm phương pháp sinh học, trong đó các hạt vàng hoặc vonfram được phủ bằng DNA và sau đó bắn vào các tế bào thực vật non,[62] và kết hợp điện, bao gồm sử dụng một cú sốc điện để làm cho màng tế bào có thể thẩm thấu DNA plasmid.

Vì chỉ có một tế bào được biến nạp vật chất di truyền, nên sinh vật phải được tái sinh từ tế bào đơn lẻ đó. Ở thực vật, điều này được thực hiện thông qua việc sử dụng nuôi cấy mô.[63][64] Ở động vật, cần đảm bảo rằng DNA được chèn vào có trong tế bào gốc phôi.[65] Vi khuẩn bao gồm một tế bào đơn lẻ và sinh sản vô tính nên việc tái tạo là không cần thiết. Các điểm đánh dấuthể lựa chọn được sử dụng để dễ dàng phân biệt các tế bào đã biến đổi với các tế bào chưa được biến đổi. Những dấu hiệu này thường có trong sinh vật chuyển gen, mặc dù một số chiến lược đã được phát triển để có thể loại bỏ dấu hiệu có thể lựa chọn khỏi cây chuyển gen trưởng thành.[66]

Thử nghiệm thêm bằng cách sử dụng PCR, lai phương Nam và giải trình tự DNA được tiến hành để xác nhận rằng một sinh vật có chứa gen mới.[67] Các xét nghiệm này cũng có thể xác nhận vị trí nhiễm sắc thể và số lượng bản sao của gen được chèn vào. Sự hiện diện của gen không đảm bảo nó sẽ được biểu hiện ở các mức độ thích hợp trong mô đích, vì vậy các phương pháp tìm kiếm và đo lường các sản phẩm của gen (RNA và protein) cũng được sử dụng. Chúng bao gồm lai phía bắc, định lượng RT-PCR, Western blot, miễn dịch huỳnh quang, ELISA và phân tích kiểu hình.[68]

Vật liệu di truyền mới có thể được đưa vào ngẫu nhiên trong bộ gen của vật chủ hoặc được nhắm mục tiêu đến một vị trí cụ thể. Kỹ thuật nhắm mục tiêu gen sử dụng tái tổ hợp tương đồng để tạo ra những thay đổi mong muốn đối với một gen nội sinh cụ thể. Điều này có xu hướng xảy ra với tần suất tương đối thấp ở thực vật và động vật và thường yêu cầu sử dụng các dấu hiệu có thể lựa chọn. Tần suất nhắm mục tiêu gen có thể được tăng cường đáng kể thông qua chỉnh sửa bộ gen. Chỉnh sửa bộ gen sử dụng các nucleaza được thiết kế nhân tạo để tạo ra các đứt gãy sợi kép cụ thể tại các vị trí mong muốn trong bộ gen và sử dụng các cơ chế nội sinh của tế bào để sửa chữa sự đứt gãy do các quá trình tự nhiên của quá trình tái tổ hợp tương đồngliên kết cuối không giống nhau. Có bốn họ nucleaza được thiết kế: meganucleaza,[69][70] nucleaza ngón tay kẽm,[71][72] nucleaza hiệu ứng giống chất hoạt hóa phiên mã (TALENs),[73][74] và hệ thống Cas9-guideRNA (đã điều chỉnh từ CRISPR).[75][76] TALEN và CRISPR là hai loại được sử dụng phổ biến nhất và mỗi loại đều có những ưu điểm riêng.[77] TALENs có độ đặc hiệu mục tiêu cao hơn, trong khi CRISPR dễ thiết kế hơn và hiệu quả hơn.[77] Ngoài việc tăng cường khả năng nhắm mục tiêu gen, các nucleaza đã được thiết kế có thể được sử dụng để tạo ra các đột biến tại các gen nội sinh tạo ra loại gen.[78][79]

Ứng dụng

Kỹ thuật di truyền có ứng dụng trong y học, nghiên cứu, công nghiệp và nông nghiệp và có thể được sử dụng trên nhiều loại thực vật, động vật và vi sinh vật. Vi khuẩn, những sinh vật đầu tiên được biến đổi gen, có thể được chèn DNA plasmid chứa các gen mới mã hóa thuốc hoặc enzym chế biến thực phẩm và các chất nền khác.[80][81] Thực vật đã được biến đổi để bảo vệ côn trùng, kháng thuốc diệt cỏ, kháng vi rút, tăng cường dinh dưỡng, khả năng chịu áp lực môi trường và sản xuất vắc xin ăn được.[82] Hầu hết các GMO được thương mại hóa là cây trồng kháng sâu bọ hoặc kháng thuốc diệt cỏ.[83] Động vật biến đổi gen đã được sử dụng để nghiên cứu, động vật mô hình và sản xuất các sản phẩm nông nghiệp hoặc dược phẩm. Động vật biến đổi gen bao gồm động vật có gen bị loại bỏ, tăng tính nhạy cảm với bệnh tật, kích thích tố để tăng trưởng thêm và khả năng thể hiện protein trong sữa của chúng.[84]

Y học

Kỹ thuật di truyền có nhiều ứng dụng trong y học bao gồm sản xuất thuốc, tạo ra động vật mô hình bắt chước tình trạng của con người và liệu pháp gen. Một trong những ứng dụng đầu tiên của kỹ thuật di truyền là sản xuất hàng loạt insulin người ở vi khuẩn.[32] Ứng dụng này hiện đã được áp dụng cho, hormone tăng trưởng ở người, hormone kích thích nang trứng (để điều trị vô sinh), albumin người, kháng thể đơn dòng, các yếu tố chống bệnh máu khó đông, vắc xin và nhiều loại thuốc khác.[85][86] Các tế bào lai chuột, các tế bào hợp nhất với nhau để tạo ra kháng thể đơn dòng, đã được điều chỉnh thông qua kỹ thuật di truyền để tạo ra kháng thể đơn dòng của người.[87] Vào năm 2017, kỹ thuật di truyền của các thụ thể kháng nguyên chimeric trên tế bào T của chính bệnh nhân đã được FDA Hoa Kỳ phê duyệt như một phương pháp điều trị ung thư bạch cầu nguyên bào lympho cấp tính. Các vi rút biến đổi gen đang được phát triển vẫn có thể tạo ra khả năng miễn dịch, nhưng thiếu các trình tự lây nhiễm.[88]

Kỹ thuật di truyền cũng được sử dụng để tạo ra các mô hình động vật về bệnh ở người. Chuột biến đổi gen là mô hình động vật biến đổi gen phổ biến nhất.[89] Chúng đã được sử dụng để nghiên cứu và mô hình hóa ung thư (ung thư tại chỗ), béo phì, bệnh tim, tiểu đường, viêm khớp, lạm dụng chất kích thích, lo lắng, lão hóa và bệnh Parkinson.[90] Có thể thử nghiệm các phương pháp chữa trị tiềm năng đối với các mẫu chuột này. Ngoài ra, lợn biến đổi gen cũng đã được lai tạo với mục đích tăng khả năng thành công của việc ghép tạng từ lợn sang người.[91]

Liệu pháp gen là kỹ thuật di truyền của con người, nói chung bằng cách thay thế các gen bị lỗi bằng các gen hiệu quả. Nghiên cứu lâm sàng sử dụng liệu pháp gen soma đã được tiến hành với một số bệnh, bao gồm SCID liên kết X,[92] bệnh bạch cầu lymphocytic mãn tính (CLL),[93][94]bệnh Parkinson.[95] Năm 2012, Alipogene tiparvovec trở thành liệu pháp điều trị gen đầu tiên được chấp thuận sử dụng trong lâm sàng.[96][97] Vào năm 2015, một loại vi-rút đã được sử dụng để chèn một gen khỏe mạnh vào tế bào da của một cậu bé mắc một chứng bệnh về da hiếm gặp, epidermolysis bullosa, để phát triển, và sau đó ghép da khỏe mạnh lên 80% cơ thể cậu bé bị ảnh hưởng do chứng bệnh này.[98]

Liệu pháp gen Germline sẽ cho kết quả trong bất kỳ sự thay đổi là được thừa kế, mà đã dấy lên lo ngại trong cộng đồng khoa học.[99][100] Vào năm 2015, CRISPR được sử dụng để chỉnh sửa DNA của phôi người không thể sống được,[101][102] các nhà khoa học hàng đầu của các học viện lớn trên thế giới kêu gọi tạm hoãn chỉnh sửa bộ gen người có thể thừa kế.[103] Cũng có những lo ngại rằng công nghệ này có thể được sử dụng không chỉ để điều trị mà còn để tăng cường, sửa đổi hoặc thay đổi ngoại hình, khả năng thích ứng, trí thông minh, tính cách hoặc hành vi của con người.[104] Sự phân biệt giữa chữa bệnh và tăng cường cũng có thể khó thiết lập.[105] Vào tháng 11 năm 2018, He Jiankui thông báo rằng anh ta đã chỉnh sửa bộ gen của hai phôi thai người, để cố gắng vô hiệu hóa gen CCR5, gen mã hóa cho một thụ thể mà HIV sử dụng để xâm nhập vào tế bào. Anh ấy nói rằng hai bé gái sinh đôi, Lulu và Nana, đã chào đời vài tuần trước đó. Ông nói rằng các cô gái vẫn mang các bản sao chức năng của CCR5 cùng với CCR5 khuyết tật (bệnh khảm) và vẫn dễ bị nhiễm HIV. Công việc này bị nhiều người lên án là phi đạo đức, nguy hiểm và quá sớm.[106]

Các nhà nghiên cứu đang thay đổi bộ gen của lợn để tạo ra sự phát triển của các bộ phận cơ thể người dùng trong cấy ghép. Các nhà khoa học đang tạo ra "ổ gen", thay đổi bộ gen của muỗi để khiến chúng miễn dịch với bệnh sốt rét, và sau đó tìm cách truyền bá muỗi đã biến đổi gen trong quần thể muỗi với hy vọng loại bỏ căn bệnh này.[107]

Nghiên cứu

Kỹ thuật gen là một công cụ quan trọng đối với các nhà khoa học tự nhiên, với việc tạo ra các sinh vật chuyển gen là một trong những công cụ quan trọng nhất để phân tích chức năng gen.[108] Các gen và thông tin di truyền khác từ nhiều loại sinh vật có thể được đưa vào vi khuẩn để lưu trữ và sửa đổi, tạo ra vi khuẩn biến đổi gen trong quá trình này. Vi khuẩn rẻ, dễ phát triển, vô tính, nhân lên nhanh chóng, tương đối dễ biến nạp và có thể bảo quản ở -80   °C gần như vô hạn. Khi một gen được phân lập, nó có thể được lưu trữ bên trong vi khuẩn, cung cấp nguồn cung cấp không giới hạn cho nghiên cứu.[109] Các sinh vật được biến đổi gen để khám phá các chức năng của một số gen nhất định. Điều này có thể là ảnh hưởng đến kiểu hình của sinh vật, nơi gen được biểu hiện hoặc những gen khác mà nó tương tác. Các thí nghiệm này thường liên quan đến việc mất chức năng, tăng chức năng, theo dõi và biểu hiện.

  • Các thí nghiệm mất chức năng, chẳng hạn như trong một thí nghiệm loại bỏ gen, trong đó một sinh vật được thiết kế để thiếu hoạt động của một hoặc nhiều gen. Trong một lần loại trực tiếp đơn giản, một bản sao của gen mong muốn đã bị thay đổi để khiến nó không hoạt động. Tế bào gốc phôi kết hợp gen đã thay đổi, gen này thay thế bản sao chức năng đã có. Các tế bào gốc này được tiêm vào phôi nang, chúng sẽ được cấy vào các bà mẹ mang thai. Điều này cho phép người thí nghiệm phân tích các khiếm khuyết do đột biến này gây ra và từ đó xác định vai trò của các gen cụ thể. Nó được sử dụng đặc biệt thường xuyên trong sinh học phát triển.[110] Khi điều này được thực hiện bằng cách tạo ra một thư viện các gen có đột biến điểm ở mọi vị trí trong vùng quan tâm, hoặc thậm chí mọi vị trí trong toàn bộ gen, điều này được gọi là "quét đột biến". Phương pháp đơn giản nhất, và là phương pháp đầu tiên được sử dụng, là "quét alanin", trong đó mọi vị trí lần lượt bị đột biến thành axit amin không hoạt động là alanin.[111]
  • Đạt được các thử nghiệm chức năng, đối chứng hợp lý của loại trực tiếp. Đôi khi chúng được thực hiện cùng với các thí nghiệm loại trực tiếp để thiết lập chức năng của gen mong muốn một cách chính xác hơn. Quá trình này tương tự như trong kỹ thuật loại trực tiếp, ngoại trừ cấu trúc được thiết kế để tăng chức năng của gen, thường bằng cách cung cấp thêm các bản sao của gen hoặc tạo ra sự tổng hợp protein thường xuyên hơn. Tăng chức năng được sử dụng để cho biết liệu một protein có đủ cho một chức năng hay không, nhưng không phải lúc nào cũng có nghĩa là nó cần thiết, đặc biệt là khi giải quyết vấn đề dư thừa chức năng hoặc di truyền.[110]
  • Theo dõi các thí nghiệm, nhằm thu thập thông tin về vị trí và sự tương tác của protein mong muốn. Một cách để làm điều này là thay thế gen kiểu hoang dã bằng gen 'dung hợp', là sự kết hợp của gen kiểu hoang dã với một phần tử báo cáo như protein huỳnh quang xanh (GFP) sẽ cho phép dễ dàng hình dung các sản phẩm của việc chỉnh sửa gen. Mặc dù đây là một kỹ thuật hữu ích, nhưng thao tác này có thể phá hủy chức năng của gen, tạo ra các hiệu ứng thứ cấp và có thể gây nghi ngờ về kết quả của thí nghiệm. Các kỹ thuật phức tạp hơn hiện đang được phát triển có thể theo dõi các sản phẩm protein mà không làm giảm chức năng của chúng, chẳng hạn như việc bổ sung các trình tự nhỏ sẽ đóng vai trò là mô-típ liên kết với các kháng thể đơn dòng.[110]
  • Các nghiên cứu về biểu hiện nhằm khám phá nơi và khi nào các protein cụ thể được sản xuất. Trong các thí nghiệm này, trình tự DNA trước DNA mã hóa một protein, được gọi là trình tự khởi động của gen, được đưa vào một sinh vật có vùng mã hóa protein được thay thế bằng một gen báo cáo như GFP hoặc một enzym xúc tác sản xuất thuốc nhuộm.. Do đó có thể quan sát được thời gian và địa điểm nơi một loại protein cụ thể được sản xuất. Các nghiên cứu biểu hiện có thể được thực hiện một bước xa hơn bằng cách thay đổi trình tự khởi động để tìm ra những phần nào là quan trọng cho sự biểu hiện thích hợp của gen và thực sự được liên kết bởi các protein yếu tố phiên mã; Quá trình này được gọi là promoter bashing.[112]

Công nghiệp

Sản phẩm của công nghệ gen

Các sinh vật có thể biến đổi tế bào của chúng với một gen mã hóa cho một loại protein hữu ích, chẳng hạn như một loại enzyme, để chúng biểu hiện quá mức protein mong muốn. Sau đó có thể tạo ra một lượng lớn protein bằng cách nuôi cấy sinh vật đã biến đổi trong thiết bị lò phản ứng sinh học sử dụng quá trình lên men công nghiệp, và sau đó tinh chế protein.[113] Một số gen không hoạt động tốt ở vi khuẩn, vì vậy nấm men, tế bào côn trùng hoặc tế bào động vật có vú cũng có thể được sử dụng.[114] Những kỹ thuật này được sử dụng để sản xuất các loại thuốc như insulin, hormone tăng trưởng ở ngườivắc xin, các chất bổ sung như tryptophan, hỗ trợ sản xuất thực phẩm (chymosin trong sản xuất pho mát) và nhiên liệu.[115] Các ứng dụng khác với vi khuẩn biến đổi gen có thể liên quan đến việc khiến chúng thực hiện các nhiệm vụ ngoài chu trình tự nhiên của chúng, chẳng hạn như sản xuất nhiên liệu sinh học,[116] làm sạch dầu tràn, carbon và các chất thải độc hại khác [117] và phát hiện asen trong nước uống.[118] Một số vi sinh vật biến đổi gen nhất định cũng có thể được sử dụng trong khai thác sinh họcxử lý sinh học, do khả năng chiết xuất kim loại nặng từ môi trường của chúng và kết hợp chúng thành các hợp chất dễ thu hồi hơn.[119]

Trong khoa học vật liệu, một loại virus biến đổi gen đã được sử dụng trong phòng thí nghiệm nghiên cứu như một giá đỡ để lắp ráp pin lithium-ion thân thiện với môi trường hơn.[120][121] Vi khuẩn cũng đã được thiết kế để hoạt động như cảm biến bằng cách biểu hiện một protein huỳnh quang trong các điều kiện môi trường nhất định.[122]

Nông nghiệp

Một trong những ứng dụng nổi tiếng nhất và gây tranh cãi của kỹ thuật gen là tạo ra và sử dụng cây trồng biến đổi gen hoặc vật nuôi biến đổi gen để sản xuất thực phẩm biến đổi gen . Các loại cây trồng đã được phát triển để tăng sản lượng, tăng khả năng chống chịu với các căng thẳng phi sinh học, thay đổi thành phần của thực phẩm hoặc để tạo ra các sản phẩm mới. [124]

Những cây trồng đầu tiên được công bố thương mại trên quy mô lớn đã cung cấp khả năng bảo vệ khỏi côn trùng gây hại hoặc khả năng chống chịu thuốc diệt cỏ . Các cây trồng kháng nấm và vi rút cũng đã được phát triển hoặc đang trong quá trình phát triển. [125] [126] Điều này làm cho việc quản lý côn trùng và cỏ dại của cây trồng dễ dàng hơn và có thể gián tiếp tăng năng suất cây trồng. [127] [128] Các cây trồng biến đổi gen trực tiếp cải thiện năng suất bằng cách thúc đẩy tăng trưởng hoặc làm cho cây cứng cáp hơn (bằng cách cải thiện khả năng chịu mặn, lạnh hoặc hạn hán) cũng đang được phát triển. [129] Vào năm 2016, cá hồi đã được biến đổi gen với hormone tăng trưởng để đạt kích thước trưởng thành bình thường nhanh hơn nhiều. [130]

GMO đã được phát triển để thay đổi chất lượng sản phẩm bằng cách tăng giá trị dinh dưỡng hoặc cung cấp chất lượng hoặc số lượng hữu ích hơn trong công nghiệp. [131] Khoai tây Amflora tạo ra một hỗn hợp tinh bột hữu ích hơn trong công nghiệp. Đậu nànhhạt cải đã được biến đổi gen để tạo ra nhiều loại dầu tốt cho sức khỏe. [132] [133] Thực phẩm biến đổi gen đầu tiên được thương mại hóa là cà chua có khả năng làm chậm quá trình chín, làm tăng thời hạn sử dụng . [134]

Thực vật và động vật đã được thiết kế để tạo ra những vật liệu mà chúng không thường làm ra. Dược phẩm sử dụng cây trồng và vật nuôi làm lò phản ứng sinh học để sản xuất vắc-xin, thuốc trung gian hoặc chính thuốc; sản phẩm hữu ích được tinh chế từ thu hoạch và sau đó được sử dụng trong quy trình sản xuất dược phẩm tiêu chuẩn. [135] Bò và dê đã được thiết kế để vắt thuốc và các protein khác trong sữa của chúng, và vào năm 2009, FDA đã phê duyệt một loại thuốc được sản xuất từ sữa dê. [136] [137]

Các ứng dụng khác

Kỹ thuật di truyền có những ứng dụng tiềm năng trong bảo tồn và quản lý khu vực tự nhiên. Việc chuyển gen thông qua các vectơ vi rút đã được đề xuất như một phương tiện kiểm soát các loài xâm lấn cũng như tiêm phòng các loài động vật bị đe dọa khỏi dịch bệnh. [138] Cây chuyển gen đã được đề xuất như một cách để tạo khả năng chống lại mầm bệnh trong các quần thể hoang dã. [139] Với những rủi ro ngày càng tăng của maladaptation trong các sinh vật như là kết quả của sự thay đổi khí hậu và xáo trộn khác, thích ứng tạo điều kiện thông qua tinh chỉnh gen có thể là một giải pháp để giảm thiểu rủi ro tuyệt chủng. [140] Các ứng dụng của công nghệ gen trong bảo tồn cho đến nay chủ yếu là lý thuyết và chưa được đưa vào thực tế.

Kỹ thuật di truyền cũng đang được sử dụng để tạo ra nghệ thuật vi sinh vật . [141] Một số vi khuẩn đã được biến đổi gen để tạo ra những bức ảnh đen trắng. [142] Các mặt hàng mới lạ như hoa cẩm chướng màu hoa oải hương, [143] hoa hồng xanh, [144]cá phát sáng [145] [146] cũng đã được sản xuất thông qua kỹ thuật gen.

Quy định

Quy định về công nghệ gen liên quan đến các phương pháp tiếp cận mà các chính phủ thực hiện để đánh giá và quản lý các rủi ro liên quan đến việc phát triển và phát hành GMO. Tình trạng pháp lý và quản lý của thực phẩm biến đổi gen khác nhau tùy theo quốc gia, với một số quốc gia cấm hoặc hạn chế chúng, và một số quốc gia khác cho phép với các mức độ quy định khác nhau. Tranh cãi

Các nhà phê bình đã phản đối việc sử dụng kỹ thuật di truyền trên một số lý do, bao gồm các mối quan tâm về đạo đức, sinh thái và kinh tế. [170] Các vấn đề đạo đức khác được nêu ra bao gồm cấp bằng sáng chế về sự sống, [171] việc sử dụng quyền sở hữu trí tuệ, [172] mức độ ghi nhãn trên sản phẩm, [173] [174] kiểm soát nguồn cung cấp thực phẩm [175] và tính khách quan của quy định quá trình.
